She lived in Sylvania Township and attended Catholic schools in Toledo, including the all-female Notre Dame Academy. Her first appearance was in The Ice Storm (1997). Then a videotape was sent to Kevin Williamson's "Dawson's Creek" (1998) and she won the role as Joey Potter. Dawson's Creek helped Holmes reach stardom, praised by critics and fans.

Holmes' first movie leading role came in Disturbing Behavior (1998) where she won a MTV Movie Award for Best Breakthrough Performance for the role. However, she has not found the same success in films as she did on television.

In 2005, she was engaged to marry the actor Tom Cruise, which made her the center of international press attention. Before that, she dated her Dawson's Creek co-Star Joshua Jackson for several months and was once engaged to actor Chris Klein.

On October 5, 2005, People magazine broke the news Holmes is pregnant. The child will be Holmes's first and Cruise's first biological child.
